بررسی تنوع ژنتیکی گونه‌های تمشک (.Rubus spp) در استان گلستان با استفاده از نشانگر AFLP

چکیده

چکیده
جنس تمشک (Rubus) از خانوادۀ Rosaceae حدود 750 گونه در دنیا دارد که 8 گونه و 5 دورگۀ آن از ایران گزارش شده است. پژوهش حاضر به‌منظور ارزیابی تنوع ژنتیکی 21 جمعیت از 6 گونۀ جنس Rubus در نقاط مختلف استان گلستان با استفاده از 9 ترکیب آغازگری AFLP انجام شد. نرم‌افزارهای Excel و Pop Gen به‌منظور تجزیه‌و‌تحلیل داده‌ها استفاده شدند. ماتریس تشابه و تجزیۀ خوشه‌ای با نرم‌افزار Past محاسبه و نمودار بر اساس الگوریتم و UPGMA با استفاده از ضریب تشابه جاکارد ترسیم شد. بر اساس نتایج پژوهش حاضر، 8 ترکیب (از 9 ترکیب آغازگر بررسی‌شده‌) چندشکلی مناسبی نشان دادند. از بین ترکیب‌های آغازگری، M160-E110 با مقدار 203/0، بیشترین میزان تنوع ژنتیکی را نشان داد و برای بررسی تنوع ژنتیکی بهتر است. آغازگرها در مجموع توانستند 505 مکان را شناسایی کنند که 277 مکان از آنها، چندشکلی نشان دادند.
بر اساس نتایج نمودار، جمعیت‌های مطالعه‌شده به دو گروه تقسیم شدند و جمعیت‌های ناحیۀ خزری و ایرانی - تورانی تا حدودی از جمعیت‌های متعلق به ناحیۀ خزری جدا شدند. طبق نتایج تجزیۀ ارتباط، بیشترین تعداد نشانگرها با صفت‌های طول دمبرگ، طول برگچۀ انتهایی، طول برگچۀ جانبی، طول گل‌آذین، عرض برگچۀ جانبی و طول دمگل ارتباط داشتند. نتایج پژوهش حاضر نشان دادند نشانگر AFLP کارایی زیادی در جداسازی جمعیت‌های گونه‌های بررسی‌شده از جنس Rubus دارد.

Abstract
The genus Rubus, from the family ofRosaceae Juss, has about 750 species worldwide, of which 8 species and 5 hybrids have been reported from Iran. The present study was conducted to evaluate the genetic diversity of 21 populations of 6 species of Rubus from different parts of Golestan province using 9 AFLP primer combinations. Excel and Pop Gen software were used for data analysis. Similarity matrix and cluster analysis were used by PAST software. The dendrogram was plotted based on unweighted pair group method with arithmetic average (UPGMA), using Jaccard's similarity coefficient. The results showed that eight out of nine selected primer combinations had appropriate polymorphic primer combinations. M160-E110 with 0.203 had the highest genetic variation among all used markers. All selected primer combinations found 505 position and 277 loci locations. Based on the results of the dendrogram, the studied populations were divided into two groups. Based on the results of the dendrogram, the studied populations were divided into two groups and the Caspian-Irano-Turanian populations were partially separated from the Caspian populations. According to the results of correlation analysis, the highest number of markers were related to traits of petiole length, terminal leaflet length, lateral leaf length, inflorescence length, lateral leaf width, and peduncle length. Results showed that AFLP marker was highly efficient in the separation and isolation of populations of species of Rubus species in this study.
